The cylinder doesn't rotate anymore, What do I need to do?
Have you tried removing the cylinder? You might have put it back incorrectly last time you cleaned it but just a guess on my part. That would be what I would do first.

done that the little pin that rotate the cylinder does not extended out to rotate the cylinder
Sounds like the pin has sheared off if I understand what you are talking about. I suggest you remove the cylinder pin and cylinder. Now cycle the hammer watching the little tab at the bottom. Now it will drop below the frame or flush with it on the second click or where you can rotate the cylinder. Now first, third and 4 clicks all lock the cylinder. Some guns I've seen the third click looks like it falls back to the second and if you try to rotate it then it will lock up the whole gun. My only other suggestion is to take it to a good gunsmith.

I agree with joec, take it to a gunsmith. At least he should be able to ID the problem.
